Does anybody know where PCNA is located, I can't fucking find shit (thanx, Google). Also, does anybody know how the killing effects of UV light work vs. the mutagenic effects? I'm assuming, the former causes disruptions in metabolism whereas the other one acts by causing pyrimidine dimers in DNA.

PCNA is in the nucleus of eukaryotic cells.

What do you mean by killing effects? Do you mean UV induced apoptosis via DNA damage?

Well, that's what I'd assume. That there is so much damage to the DNA that either apoptosis occurs or no amount of translesion synthesis can replicate past the amount of present dimers.

Apoptosis can occur because of substantial DNA damage, which is what happens in UV induced apoptosis.

Even if apoptosis doesn't occur and DNA damage gets through, it's possible that the damaged DNA codes for proteins vital to the proper function of cell (eg. metabolism, cell surface proteins, enzymes...) that could cause toxicity within the cells formed after mitosis. This toxicity could then cause the new cells to enter apoptosis.

Or it could cause mutations in oncogenes or tumour suppressor proteins, progressing that cell line towards an oncogenetic future.
