I am making some sprites for a game I'm developing. I've been using Paint so far to create the images, however I'm finding that sometimes paint decides to 'compress' the image while I'm working with it, causing artifacting. I need the images in JPEG format, and I need the pixels to stay at PRECISELY the values I set for them.

JPEG is a lossy format, so you get what you get. Use TGA or PNG or something lossless if you really need exact pixels.

JPEG needs to be re-encoded every time you save. Not technically, some affine transforms can be done without a need to re-encode but I have yet to find any software that will actually bother checking for this

Limiting the recompression to macroblocks is possible. I believe there was some editor around supporting this (recompress only changed macroblocks!), however editing jpg's is gay in itself. Use real lossless formats OP.
if you really need a jpeg, you should keep it in a lossless format like png for editing, and convert it to jpeg once when you're done. never open the jpeg for editing. never convert from jpeg to png.
