Save this file as "dont.bat" in your C:\Windows folder.
Then open a command prompt and type "dont delete FILE".

that only works if the person who wants to delete the file can't boot the machine from a live cd... or just put the hard drive in a different machine and delete it...

for ntfs directly modify mft with hex editor to mark certain sectors as allocated, then store data in those sectors using your own custom-written ifs driver.
if the media is writable, then of course the file is deletable.
You can either set permissions, but if someone has physical access, of course they can delete it, don't be stupid.
You can do some tricks with the filesystem that would make the OS unable to delete a file, but all the tricks can be undone of course, after all, IT IS writable media.
put it on a cd and launch it into space.
we'll probably be extinct long before that copy of the file is destroyed. assuming of course you don't launch it on an intercept course with anything in our solar system.
