The Hamza is not a vowel. But the sukun is.
The difference is rather important I wish I knew how to explain it to you :(
Hamza is like the sound between o and i in going whereas sukun simply states that the letter is followed by another letter without an a i or u, in between.
If you think that is nonsense keep in mind that the short vowels are actually not written at all but are only used in the Qur'an, poems and books that teach arabic.

The sign you're talking about is called hamza and it represents a consonant sound that doesn't exist in English. It's made by closing together your vocal chords and releasing, kinda like the sound of a very mild dry-heave.
