On Windows XP I have two drives, how do I block the second drive (not the System) from being accessed by the Guest account?

Disable simple file sharing and all your root files from that drive and put them all in one folder, revoke guest or whichever account access to folder. If you dont want users to write on the root directory--add disk quota management make it 1 KB (thats the minimum filesize you can get at, obviously... anything they can write is insignificant)

Try right clicking the empty space on your drive's root directory and deny all rights for Guest account.
